چکیده

87 Progress in fluorescence imaging is enabling the study of biological events at unprecedented detail, thanks to novel microscopy techniques that provide imaging with nanoscale resolution. 3D imaging of living cells within intact tissues, organs, and whole animals, is accessible through multiphoton excitation. The remarkably rich photophysics of Fluorescent Proteins (FPs) makes them flexible enough to be used in conjunction with these advanced spectroscopic approaches. Here we report on our progress in understanding FPs multiphoton excitation spectra and in unraveling the photophysics behind photoactivation/deactivation of some FPs, a property enabling their use as fluorophores for nano-resolution imaging. This analysis allows us to develop new reversibly switchable FP mutants.

Simultaneous imaging of GFP, CFP and collagen in tumors in vivo using multiphoton microscopy

BACKGROUND The development of multiphoton laser scanning microscopy has greatly facilitated the imaging of living tissues. However, the use of genetically encoded fluorescent proteins to distinguish different cell types in living animals has not been described at single cell resolution using multiphoton microscopy. RESULTS Here we describe a method for the simultaneous imaging, by multiphoton...

Advanced Methods in Fluorescence Microscopy

It requires a good deal of will power to resist hyperbole in considering the advances that have been achieved in fluorescence microscopy in the last 25 years. Our effort has been to survey the modalities of microscopic fluorescence imaging available to cell biologists and perhaps useful for diagnostic pathologists. The gamut extends from established confocal laser scanning through multiphoton a...

Multiphoton microscopy of cleared mouse brain expressing YFP.

Multiphoton microscopy of intrinsic fluorescence and second harmonic generation (SHG) of whole mouse organs is made possible by optically clearing the organ before imaging.(1,2) However, for organs that contain fluorescent proteins such as GFP and YFP, optical clearing protocols that use methanol dehydration and clear using benzyl alcohol:benzyl benzoate (BABB) while unprotected from light(3) d...

Genetic Code Expansion Enables Live-Cell and Super-Resolution Imaging of Site-Specifically Labeled Cellular Proteins

Methods to site-specifically and densely label proteins in cellular ultrastructures with small, bright, and photostable fluorophores would substantially advance super-resolution imaging. Recent advances in genetic code expansion and bioorthogonal chemistry have enabled the site-specific labeling of proteins. However, the efficient incorporation of unnatural amino acids into proteins and the spe...

Cell-based and in vivo spectral analysis of fluorescent proteins for multiphoton microscopy.

Multiphoton microscopy of cells and subcellular structures labeled with fluorescent proteins is the state-of-the-art technology for longitudinal imaging studies in tissues and living animals. Successful analysis of separate cell populations or signaling events by intravital microscopy requires optimal pairing of multiphoton excitation wavelengths with spectrally distinct fluorescent proteins. W...
